AOS as stated is the newest version of UO, allowing players to use power scroll to raise their skill cap levels, having items and weapons that allow them to regain med quicker, hit faster etc etc.... Diablo style clone to be honest. On AOS you really do need items to pvp, but most people agree that player versus monster is more fun on AOS too, since not only tamers can do it.
UO:R is post 99 UO up until the release of AOS. It is the time where the poison/harm combo existed, and required quite a good con to stay competitive in the PvP, and from this era on you must cure poison before healing. It does however offer a good number of PvP templates including: tank mages/healer mages/scribe+poison mages/lumberjack warriors/each dexer skill has 'special blows'. It does seem the most popular era of UO for most people. UO:R has slower styled pvp (spells cast slower, run out of mana etc), but there is so much more teamwork there that makes it fun.
T2A this is post 98 - pre 99. This is for more of an old UO feel to it. There is no neon items or special items here, just good old fashioned UO. The PvP isn't as varied as UO:R, with the tank mage dominating PvP, though some do still play dexers. The skill and stat gains are much harder. You can heal through poison on this era, and it also features the old 'instant hit'. The shard is much tougher in a lot of aspects, harder to gain gold, the public moongates don't have a menu system... however this is how a lot of people feel UO should be. Mainly on AOS you can reach 120 in skills, there are 2 new skills Chivalry and Necromancy, as well as a completely overhauled combat system. It's all pretty much item based, as in if your not wearing armor with resistances you die to Explosion & Flame Strike. The PvM is more interesting than UO:R as there's more places to hunt, new mobs etc. The PvP is far different than UO:R, one thing I have noticed is if your out to hunt (ie wearing luck armor) you can barely defend yourself against a player killer, fights don't last nearly as long. All and all it's fun to play for a little while, but since it's so item based, you will have to farm items to pvp.

A great difference is the addition of two new skills: Chivalry and Necromancy,
And two new facets where you can live, Malas and Ilshenar.
Also Spirit Speak has been improved.
